Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.1881-2A>C, citing Ambry Variant Classification Scheme 2023: The c.1881-2A>C intronic variant results from an A to C substitution two nucleotides upstream from coding exon 16 in the EGFR gene. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. A resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNAdecay; although, direct evidence is unavailable. This nucleotide position is highly conserved in available vertebrate species. Based on the available evidence, the clinical significance of this variant remains unclear.
