Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003977.4(AIP):c.88G>A (p.Asp30Asn), citing Ambry Variant Classification Scheme 2023: The p.D30N variant (also known as c.88G>A), located in coding exon 1 of the AIP gene, results from a G to A substitution at nucleotide position 88. The aspartic acid at codon 30 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
