NM_000075.4(CDK4):c.308A>C (p.Tyr103Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y103S variant (also known as c.308A>C), located in coding exon 2 of the CDK4 gene, results from an A to C substitution at nucleotide position 308. The tyrosine at codon 103 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
