This variant lies in the HADHA gene (transcript NM_000182.5) at coding-DNA position 2229 through coding-DNA position 2233, duplicating 5 bases; at the protein level this means shifts the reading frame starting at threonine residue 745, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of long-chain 3-hydroxyacyl-CoA dehydrogenase deficiency (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Thr745Serfs*8) in the HADHA gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 19 amino acid(s) of the HADHA protein.
