NM_001042492.3(NF1):c.4865T>G (p.Leu1622Trp) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1601W variant (also known as c.4802T>G), located in coding exon 36 of the NF1 gene, results from a T to G substitution at nucleotide position 4802. The leucine at codon 1601 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
