NM_005228.5(EGFR):c.3020T>C (p.Met1007Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1007T variant (also known as c.3020T>C), located in coding exon 25 of the EGFR gene, results from a T to C substitution at nucleotide position 3020. The methionine at codon 1007 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
