Uncertain significance for Niemann-Pick disease, type C1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000271.5(NPC1):c.3436G>A (p.Gly1146Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1146 of the NPC1 protein (p.Gly1146Ser). This variant is present in population databases (rs780006718, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with NPC1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
