Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127671.2(LIFR):c.2744C>T (p.Ser915Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with leucine at codon 915 of the LIFR protein (p.Ser915Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with LIFR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15").
