Pathogenic for Nephronophthisis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001023570.4(IQCB1):c.1532_1536dup (p.Gln513Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.01%). For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the IQCB1 protein in which other variant(s) (p.Gln520*) have been determined to be pathogenic (PMID: 26766544, 29068479, 32531858). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This premature translational stop signal has been observed in individual(s) with leber congenital amaurosis (PMID: 20881296). This sequence change creates a premature translational stop signal (p.Gln513*) in the IQCB1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 86 amino acid(s) of the IQCB1 protein.
