Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.998A>G (p.Lys333Arg), citing Ambry Variant Classification Scheme 2023: The p.K333R variant (also known as c.998A>G), located in coding exon 7 of the PTCH1 gene, results from an A to G substitution at nucleotide position 998. The lysine at codon 333 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
