Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374736.1(DST):c.4736A>G (p.Gln1579Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 1042 of the DST protein (p.Gln1042Arg). This variant is present in population databases (rs773629086, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with DST-related conditions.
