NM_017802.4(DNAAF5):c.1180G>A (p.Ala394Thr) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1180G>A (p.A394T) alteration is located in exon 5 (coding exon 5) of the DNAAF5 gene. This alteration results from a G to A substitution at nucleotide position 1180, causing the alanine (A) at amino acid position 394 to be replaced by a threonine (T). Based on data from gnomAD, the A allele has an overall frequency of 0.002% (5/250228) total alleles studied. The highest observed frequency was 0.006% (1/16198) of African alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
