Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005689.4(ABCB6):c.2167C>T (p.Arg723Trp), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 723 of the ABCB6 protein (p.Arg723Trp). This variant is present in population databases (rs199515171, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with ABCB6-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
