Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.1857C>G (p.Phe619Leu), citing Ambry Variant Classification Scheme 2023: The p.F619L variant (also known as c.1857C>G), located in coding exon 6 of the BLM gene, results from a C to G substitution at nucleotide position 1857. The phenylalanine at codon 619 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
