Uncertain significance for Ehlers-Danlos syndrome, type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000090.4(COL3A1):c.2236C>T (p.Pro746Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 746 of the COL3A1 protein (p.Pro746Ser). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt COL3A1 protein function. This missense change has been observed in individual(s) with Ehlers-Danlos syndrome (Invitae). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency).
