Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006445.4(PRPF8):c.1897G>A (p.Gly633Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 633 of the PRPF8 protein (p.Gly633Ser). This variant is present in population databases (rs753143915, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with PRPF8-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
