Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001563.4(IMPG1):c.1543_1544dup (p.Met515fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with IMPG1-related conditions. This sequence change creates a premature translational stop signal (p.Met515Ilefs*6) in the IMPG1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in IMPG1 are known to be pathogenic (PMID: 23993198). This variant is not present in population databases (gnomAD no frequency).
