Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000372.5(TYR):c.767A>G (p.His256Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 256 of the TYR protein (p.His256Arg). This variant is present in population databases (no rsID available, gnomAD 0.006%). This missense change has been observed in individual(s) with oculocutaneous albinism (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
