Pathogenic for FG syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005120.3(MED12):c.1924_1974+17dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MED12 gene (transcript NM_005120.3) at coding-DNA position 1924 through 17 bases into the intron immediately after coding-DNA position 1974, duplicating this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has been observed in individual(s) with clinical features of MED12-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 13 of the MED12 gene. It does not directly change the encoded amino acid sequence of the MED12 protein.
