This variant lies in the COL5A2 gene (transcript NM_000393.5) at coding-DNA position 3209, where G is replaced by A; at the protein level this means replaces arginine at residue 1070 with histidine — a missense variant. Submitter rationale: Variant summary: COL5A2 c.3209G>A (p.Arg1070His) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 6.8e-05 in 251294 control chromosomes. The observed variant frequency exceeds the estimated maximal expected allele frequency for disease-causing variants in COL5A2. c.3209G>A has been observed in an study on congenital heart disease (Jin_2017). These report(s) do not provide unequivocal conclusions about association of the variant with Ehlers-Danlos Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
