Uncertain significance for Hyper-IgM syndrome type 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080911.3(UNG):c.77C>A (p.Pro26Gln), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces proline, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 26 of the UNG protein (p.Pro26Gln). This variant has not been reported in the literature in individuals affected with UNG-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
