Uncertain significance for Glycogen storage disease due to muscle beta-enolase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_053013.4(ENO3):c.94C>T (p.Arg32Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg32*) in the ENO3 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ENO3 cause disease. This variant is present in population databases (rs764120380, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with ENO3-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
