Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001385641.1(SAMD11):c.934A>T (p.Arg312Ter), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with SAMD11-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change creates a premature translational stop signal (p.Arg133*) in the SAMD11 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SAMD11 cause disease.
