Uncertain significance for Epilepsy, X-linked 1, with variable learning disabilities and behavior disorders — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006950.3(SYN1):c.185C>T (p.Ser62Phe), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with SYN1-related conditions. This variant is present in population databases (rs776054489, gnomAD 0.03%). This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 62 of the SYN1 protein (p.Ser62Phe). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
