Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4154A>G (p.Tyr1385Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1385C variant (also known as c.4154A>G), located in coding exon 21 of the DICER1 gene, results from an A to G substitution at nucleotide position 4154. The tyrosine at codon 1385 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
