Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020232.5(PSMG2):c.16G>C (p.Gly6Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with PSMG2-related conditions. This variant is present in population databases (rs756591475, gnomAD 0.0009%). This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 6 of the PSMG2 protein (p.Gly6Arg).
