This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 4746, where A is replaced by G; at the protein level this means the protein sequence is unchanged (threonine at residue 1582 retained) — a synonymous variant. Submitter rationale: Variant summary: FBN1 c.4746A>G (p.Thr1582Thr) alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 251218 control chromosomes (gnomAD).
