Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 0.00095 in 238502 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for disease-causing variants in PLEC, allowing no conclusion about variant significance. c.9115G>A has been observed in individuals affected with late-onset rod-cone dystrophy or primary immunodeficiency disorders (Mjcase_2016, Schlechter_2017). These reports do not provide unequivocal conclusions about association of the variant with Epidermolysis bullosa simplex 5C, with pyloric atresia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
