Submitter rationale: The G3609S variant in the PLEC gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G3609S variant was not observed with any significant frequency in approximately 6,300 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G3609S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved, and in silico analysis predicts this variant likely does not alter the protein structure/function. We interpret G3609S as a variant of uncertain significance.
