NM_000116.5(TAFAZZIN):c.370+2T>A was classified as Uncertain significance for 3-Methylglutaconic aciduria type 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 4 of the TAZ gene. Alternative splicing of the TAZ gene results in a functional isoform lacking exon 5 (delta5) in multiple human tissues (PMID: 19619503, 24342716, 12930833, 19700766). However, it is currently unclear if variants that occur in this region of the gene cause disease. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TAZ-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
