Uncertain significance for Aortic aneurysm, familial thoracic 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002474.3(MYH11):c.1862A>G (p.Asp621Gly), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with MYH11-related conditions. This variant is present in population databases (rs773829617, gnomAD 0.0009%). This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 628 of the MYH11 protein (p.Asp628Gly). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
