Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.615G>T (p.Glu205Asp), citing Ambry Variant Classification Scheme 2023: The p.E205D variant (also known as c.615G>T), located in coding exon 5 of the RECQL gene, results from a G to T substitution at nucleotide position 615. The glutamic acid at codon 205 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
