NM_002180.3(IGHMBP2):c.660A>T (p.Lys220Asn) was classified as Uncertain significance for Autosomal recessive distal spinal muscular atrophy 1; Charcot-Marie-Tooth disease axonal type 2S by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 220 of the IGHMBP2 protein (p.Lys220Asn). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of distal hereditary motor neuropathy (PMID: 25326637, 26633542, 30755392). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
