Uncertain significance for Rienhoff syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003239.5(TGFB3):c.293C>G (p.Ser98Trp), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15". The tryptophan amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This sequence change replaces serine, which is neutral and polar, with tryptophan, which is neutral and slightly polar, at codon 98 of the TGFB3 protein (p.Ser98Trp). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with TGFB3-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
