Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001379270.1(CNGA1):c.965G>A (p.Gly322Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 326 of the CNGA1 protein (p.Gly326Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with retinitis pigmentosa (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
