Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005726.6(TSFM):c.320T>C (p.Ile107Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with threonine at codon 107 of the TSFM protein (p.Ile107Thr). The isoleucine residue is moderately conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is present in population databases (rs758156753, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with TSFM-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
