NM_000343.4(SLC5A1):c.16T>C (p.Trp6Arg) was classified as Uncertain significance for Congenital glucose-galactose malabsorption by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 6 of the SLC5A1 protein (p.Trp6Arg). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with SLC5A1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
