Pathogenic for Jeune thoracic dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020800.3(IFT80):c.28G>T (p.Glu10Ter), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This sequence change creates a premature translational stop signal (p.Glu10*) in the IFT80 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in IFT80 are known to be pathogenic (PMID: 21227999, 23339108, 29068549). This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with IFT80-related conditions. For these reasons, this variant has been classified as Pathogenic.
