This variant lies in the KIF1A gene (transcript NM_001244008.2) at coding-DNA position 710, where C is replaced by T; at the protein level this means replaces threonine at residue 237 with isoleucine — a missense variant. Submitter rationale: Variant summary: KIF1A c.710C>T (p.Thr237Ile) results in a non-conservative amino acid change located in the Kinesin motor domain (IPR001752) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249432 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.710C>T in individuals affected with NESCAV Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as uncertain significance.
