NM_003000.3(SDHB):c.134A>G (p.Tyr45Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y45C variant (also known as c.134A>G), located in coding exon 2 of the SDHB gene, results from an A to G substitution at nucleotide position 134. The tyrosine at codon 45 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
