Uncertain significance for Autoimmune lymphoproliferative syndrome, type III caused by mutation in PRKCD — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006254.4(PRKCD):c.1797C>G (p.Ile599Met), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant is present in population databases (rs781873887, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with PRKCD-related conditions. This sequence change replaces isoleucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 599 of the PRKCD protein (p.Ile599Met).
