Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5605C>T (p.Pro1869Ser), citing Ambry Variant Classification Scheme 2023: The p.P1869S variant (also known as c.5605C>T), located in coding exon 26 of the DICER1 gene, results from a C to T substitution at nucleotide position 5605. The proline at codon 1869 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
