Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_057175.5(NAA15):c.860A>C (p.Tyr287Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with serine, which is neutral and polar, at codon 287 of the NAA15 protein (p.Tyr287Ser). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with NAA15-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
