Uncertain significance for SLC35A2-congenital disorder of glycosylation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005660.3(SLC35A2):c.940C>T (p.Arg314Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 314 of the SLC35A2 protein (p.Arg314Cys). This variant is present in population databases (rs781992603, gnomAD 0.006%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with SLC35A2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1951228). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt SLC35A2 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
