Uncertain significance for Immunodeficiency 18 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000733.4(CD3E):c.71G>T (p.Gly24Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 24 of the CD3E protein (p.Gly24Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CD3E-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
