Pathogenic — the classification assigned by GeneDx to NM_022369.4(STRA6):c.1385del (p.Asn462fs), citing GeneDx Variant Classification (06012015). This variant lies in the STRA6 gene (transcript NM_022369.4) at coding-DNA position 1385, deleting one base; at the protein level this means shifts the reading frame starting at asparagine residue 462, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1385delA variant in the STRA6 gene causes a frameshift starting with codon Asparagine 462, changes this amino acid to a Threonine residue and creates a premature Stop codon at position 16 of the new reading frame, denoted p.Asn462ThrfsX16. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this variant have not been reported previously to our knowledge, we interpret it as pathogenic.
