Uncertain significance for Osteogenesis imperfecta type 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022356.4(P3H1):c.586G>A (p.Asp196Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 196 of the P3H1 protein (p.Asp196Asn). This variant is present in population databases (rs777460970, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with P3H1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
