Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003906.5(MCM3AP):c.888C>A (p.Asp296Glu), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This variant has not been reported in the literature in individuals affected with MCM3AP-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 296 of the MCM3AP protein (p.Asp296Glu). This variant is not present in population databases (gnomAD no frequency).
