Uncertain significance for Walker-Warburg congenital muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001079802.2(FKTN):c.751A>G (p.Arg251Gly), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C65"). This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 251 of the FKTN protein (p.Arg251Gly). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FKTN-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
