Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003321.5(TUFM):c.906C>G (p.Ile302Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 302 of the TUFM protein (p.Ile302Met). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TUFM-related conditions. ClinVar contains an entry for this variant (Variation ID: 1938839). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TUFM protein function with a negative predictive value of 80%. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
